    Get PDF
&nbsp

Targeted Capture of Homoeologous Coding and Noncoding Sequence in Polyploid Cotton

Targeted sequence capture is a promising technology in many areas in biology. These methods enable efficient and relatively inexpensive sequencing of hundreds to thousands of genes or genomic regions from many more individuals than is practical using whole-genome sequencing approaches. Here, we demonstrate the feasibility of target enrichment using sequence capture in polyploid cotton. To capture and sequence both members of each gene pair (homeologs) of wild and domesticated Gossypium hirsutum, we created custom hybridization probes to target 1000 genes (500 pairs of homeologs) using information from the cotton transcriptome. Two widely divergent samples of G. hirsutum were hybridized to four custom NimbleGen capture arrays containing probes for targeted genes. We show that the two coresident homeologs in the allopolyploid nucleus were efficiently captured with high coverage. The capture efficiency was similar between the two accessions and independent of whether the samples were multiplexed. A significant amount of flanking, nontargeted sequence (untranslated regions and introns) was also captured and sequenced along with the targeted exons. Intraindividual heterozygosity is low in both wild and cultivated Upland cotton, as expected from the high level of inbreeding in natural G. hirsutum and bottlenecks accompanying domestication. In addition, levels of heterozygosity appeared asymmetrical with respect to genome (A(T) or D(T)) in cultivated cotton. The approach used here is general, scalable, and may be adapted for many different research inquiries involving polyploid plant genomes

Is communication guidance mistaken? Qualitative study of parent-oncologist communication in childhood cancer

    Get PDF
background: Guidance encourages oncologists to engage patients and relatives in discussing the emotions that accompany cancer diagnosis and treatment. We investigated the perspectives of parents of children with leukaemia on the role of paediatric oncologists in such discussion. methods: Qualitative study comprising 33 audio-recorded parent–oncologist consultations and semi-structured interviews with 67 parents during the year following diagnosis. results: Consultations soon after the diagnosis were largely devoid of overt discussion of parental emotion. Interviewed parents did not describe a need for such discussion. They spoke of being comforted by oncologists’ clinical focus, by the biomedical information they provided and by their calmness and constancy. When we explicitly asked parents 1 year later about the oncologists’ role in emotional support, they overwhelmingly told us that they did not want to discuss their feelings with oncologists. They wanted to preserve the oncologists’ focus on their child’s clinical care, deprecated anything that diverted from this and spoke of the value of boundaries in the parent–oncologist relationship. conclusion: Parents were usually comforted by oncologists, but this was not achieved in the way suggested by communication guidance. Communication guidance would benefit from an enhanced understanding of how emotional support is experienced by those who rely on it

DIAGNOSIS OF TEACHING HANDBALL AT THE BASIC LEVEL OF PUBLIC EDUCATIONAL CENTERS IN CANTON MANTA ABSTRACT Playing is part of the individual's own nature. Its importance through sports practice has allowed it to become a key instrument within practice in current pedagogy, which considers it a structural element in the teaching-learning interaction. Handball is a sport that, on a cognitive level, generates in those who practice it the necessary conditions of response to the choice of technical elements and tactical procedures in the search for solutions that the game deserves, for which it is attributed the equitable development that integrates the psychic and the motor. In Educational District 13D02, province of Manabí, Ecuador, teachers in the physical education area execute the curriculum proposed by the Ministry of Education with all its pedagogical tools, however, this article aims to diagnose the current state of the teaching handball in the fiscal institutions of the Manta canton through the application of a research instrument, consisting of a bank of questions with options designed on the Likert scale, applied to the 103 teachers in the area of physical education, which allowed the systematization, analysis and interpretation of the results obtained. This research, in addition to justifying the didactic use of handball, concludes that, although the current state of handball practice is limited, the promotion of this discipline will contribute in an integral way to the strengthening of the psychomotor field within the teaching / learning process. CLEAR I: Ages and Metallicities of Quiescent Galaxies at 1.0<z<1.8\mathbf{1.0 < z < 1.8} Derived from Deep Hubble Space Telescope Grism Data

We use deep \textit{Hubble Space Telescope} spectroscopy to constrain the metallicities and (\editone{light-weighted}) ages of massive (logM/M10\log M_\ast/M_\odot\gtrsim10) galaxies selected to have quiescent stellar populations at 1.0<z<1.81.0<z<1.8. The data include 12--orbit depth coverage with the WFC3/G102 grism covering \sim 8,000<λ<11,5008,000<\lambda<11,500~\AA\, at a spectral resolution of R210R\sim 210 taken as part of the CANDELS Lyman-α\alpha Emission at Reionization (CLEAR) survey. At 1.0<z<1.81.0<z<1.8, the spectra cover important stellar population features in the rest-frame optical. We simulate a suite of stellar population models at the grism resolution, fit these to the data for each galaxy, and derive posterior likelihood distributions for metallicity and age. We stack the posteriors for subgroups of galaxies in different redshift ranges that include different combinations of stellar absorption features. Our results give \editone{light-weighted ages of tz1.1=3.2±0.7t_{z \sim 1.1}= 3.2\pm 0.7~Gyr, tz1.2=2.2±0.6t_{z \sim 1.2}= 2.2\pm 0.6~Gyr, tz1.3=3.1±0.6t_{z\sim1.3}= 3.1\pm 0.6~Gyr, and tz1.6=2.0±0.6t_{z\sim1.6}= 2.0 \pm 0.6~Gyr, \editone{for galaxies at z1.1z\sim 1.1, 1.2, 1.3, and 1.6. This} implies that most of the massive quiescent galaxies at 168168\% of their stellar mass by a redshift of z>2z>2}. The posteriors give metallicities of \editone{Zz1.1=1.16±0.29Z_{z\sim1.1}=1.16 \pm 0.29~ZZ_\odot, Zz1.2=1.05±0.34Z_{z\sim1.2}=1.05 \pm 0.34~ZZ_\odot, Zz1.3=1.00±0.31Z_{z\sim1.3}=1.00 \pm 0.31~ZZ_\odot, and Zz1.6=0.95±0.39Z_{z\sim1.6}=0.95 \pm 0.39~ZZ_\odot}. This is evidence that massive galaxies had enriched rapidly to approximately Solar metallicities as early as z3z\sim3.Comment: 32 pages, 23 figures, Resubmited to ApJ after revisions in response to referee repor

ZFOURGE: Using Composite Spectral Energy Distributions to Characterize Galaxy Populations at 1<z<4

We investigate the properties of galaxies as they shut off star formation over the 4 billion years surrounding peak cosmic star formation. To do this we categorize 7000\sim7000 galaxies from 1<z<41<z<4 into 9090 groups based on the shape of their spectral energy distributions (SEDs) and build composite SEDs with R50R\sim 50 resolution. These composite SEDs show a variety of spectral shapes and also show trends in parameters such as color, mass, star formation rate, and emission line equivalent width. Using emission line equivalent widths and strength of the 4000\AA\ break, D(4000)D(4000), we categorize the composite SEDs into five classes: extreme emission line, star-forming, transitioning, post-starburst, and quiescent galaxies. The transitioning population of galaxies show modest Hα\alpha emission (EWREST40EW_{\rm REST}\sim40\AA) compared to more typical star-forming composite SEDs at log10(M/M)10.5\log_{10}(M/M_\odot)\sim10.5 (EWREST80EW_{\rm REST}\sim80\AA). Together with their smaller sizes (3 kpc vs. 4 kpc) and higher S\'ersic indices (2.7 vs. 1.5), this indicates that morphological changes initiate before the cessation of star formation. The transitional group shows a strong increase of over one dex in number density from z3z\sim3 to z1z\sim1, similar to the growth in the quiescent population, while post-starburst galaxies become rarer at z1.5z\lesssim1.5. We calculate average quenching timescales of 1.6 Gyr at z1.5z\sim1.5 and 0.9 Gyr at z2.5z\sim2.5 and conclude that a fast quenching mechanism producing post-starbursts dominated the quenching of galaxies at early times, while a slower process has become more common since z2z\sim2.Comment: Accepted for publication in The Astrophysical Journa

Repeat anterior cruciate ligament injury and return to sport in Australian soccer players after anterior cruciate ligament reconstruction with hamstring tendon autograft

    Get PDF
Background: Soccer is the most commonly played team sport in the world and a high-risk sport for anterior cruciate ligament (ACL) injury and subsequent ACL reconstruction (ACLR). Purpose: To assess the rate of further ACL injury in patients who have undergone ACLR with hamstring tendon autograft after soccer injuries in Australia and to determine factors associated with repeat ACL injury and return to soccer. Study Design: Case-control study; Level of evidence, 3. Methods: From a prospectively collected database, a series of 1000 consecutive ACLRs using hamstring autografts performed in soccer players were identified. Patients were surveyed at a minimum 5 years after reconstruction, including details of further ACL injuries to either knee, return to soccer or other sports, and psychological readiness per the Anterior Cruciate Ligament Return to Sport after Injury (ACL-RSI) scale. Results: Of the 862 participants reviewed, ACL graft rupture occurred in 85 (10%) and contralateral ACL rupture in 68 (8%) within 5 years after the reconstruction. The 5-year ACL graft survivorship was 94% for females and 88% for males. The survivorship of the contralateral ACL was 92% for males and 90% for females. When compared with those aged \u3e25 years, the odds of ACL graft rupture was increased by 4 to 5 times in those aged 19 to 25 years and 3 to 7 times in those ≤18 years. Further ACL injury to the graft or contralateral knee occurred in 44% of males aged ≤18 years. Risk factors for further ACL injury were younger age at time of surgery, male sex, and return to soccer. Graft diameter did not influence ACL graft rupture rates, and 70% of patients returned to soccer after ACLR. The mean ACL-RSI score was 59, and patients who reported more fear of reinjury on this scale were less likely to have returned to soccer. Conclusion: The prevalence of ACL graft rupture (10%) and contralateral ACL rupture (8%) was near equivalent over 5 years in this large cohort of mostly recreational Australian soccer players. ACLR with hamstring autograft is a reliable procedure, allowing 70% of patients to return to soccer in this high-risk population. Risk factors for further ACL injury are progressively younger age at time of surgery, male sex, and return to soccer. Graft diameter was not a factor in ACL graft rupture, indicating that other factors, particularly age, are of primary importance

Systematic techniques for assisting recruitment to trials (START): study protocol for embedded, randomized controlled trials

    Get PDF
BACKGROUND: Randomized controlled trials play a central role in evidence-based practice, but recruitment of participants, and retention of them once in the trial, is challenging. Moreover, there is a dearth of evidence that research teams can use to inform the development of their recruitment and retention strategies. As with other healthcare initiatives, the fairest test of the effectiveness of a recruitment strategy is a trial comparing alternatives, which for recruitment would mean embedding a recruitment trial within an ongoing host trial. Systematic reviews indicate that such studies are rare. Embedded trials are largely delivered in an ad hoc way, with interventions almost always developed in isolation and tested in the context of a single host trial, limiting their ability to contribute to a body of evidence with regard to a single recruitment intervention and to researchers working in different contexts. METHODS/DESIGN: The Systematic Techniques for Assisting Recruitment to Trials (START) program is funded by the United Kingdom Medical Research Council (MRC) Methodology Research Programme to support the routine adoption of embedded trials to test standardized recruitment interventions across ongoing host trials. To achieve this aim, the program involves three interrelated work packages: (1) methodology - to develop guidelines for the design, analysis and reporting of embedded recruitment studies; (2) interventions - to develop effective and useful recruitment interventions; and (3) implementation - to recruit host trials and test interventions through embedded studies. DISCUSSION: Successful completion of the START program will provide a model for a platform for the wider trials community to use to evaluate recruitment interventions or, potentially, other types of intervention linked to trial conduct. It will also increase the evidence base for two types of recruitment intervention. TRIAL REGISTRATION: The START protocol covers the methodology for embedded trials. Each embedded trial is registered separately or as a substudy of the host trial
